Single molecule fluorescence spectroscopy of mutants of the Discosoma red fluorescent protein DsRed

چکیده
We studied the emission of mutants of the red fluorescent protein DsRed by room temperature single molecule fluorescence spectroscopy. Bulk samples of the DsRed variant E8 show mixed green and red fluorescence of equivalent intensities individually spectrally similar to arrested green and mature red fluorescent forms of DsRed. Investigations at the single molecule level indicate that, like DsRed, E8 is not monomeric at single molecule concentrations. The entities visualized are composed of green and red emitting proteins without a fixed ratio of green to red fluorescing units. We find indications for only weak, if any, fluorescence resonance energy transfer (FRET) between red and green chromophores within one E8 entity. 2002 Elsevier Science B.V. All rights reserved.

Photophysics of DsRed, a Red Fluorescent Protein, from the Ensemble to the Single-Molecule Level
DsRed, a tetrameric fluorescent protein cloned from the Discosoma genus of coral, has shown promise as a longer-wavelength substitute for green fluorescent protein (GFP) mutants for in vivo protein labeling. Bulk and single-molecule studies of the recombinant protein revealed that the DsRed chromophore shows high stability against photobleaching as compared to GFP mutants. Stark modulation spec...
متن کاملIdentification of different emitting species in the red fluorescent protein DsRed by means of ensemble and single-molecule spectroscopy.
The photophysics and photochemistry taking place in the DsRed protein, a recently cloned red fluorescent protein from a coral of the Discosoma genus, are investigated here by means of ensemble and single-molecule time-resolved detection and spectroscopic measurements. Ensemble time-resolved data reveal that 25% of the immature green chromophores are present in tetramers containing only this imm...

Gene expression of intrinsically fluorescent proteins in biological systems offers new noninvasive windows into cellular function, but optimization of these probes relies on understanding their molecular spectroscopy, dynamics, and structure. Here, the photophysics of red fluorescent protein (dsRed) from discosoma (coral), providing desired longer emission/absorption wavelengths, and an improve...
متن کاملThe nature of fluorescence emission in the red fluorescent protein DsRed, revealed by single-molecule detection.
Recent studies on the newly cloned red fluorescence protein DsRed from the Discosoma genus have shown its tremendous advantages: bright red fluorescence and high resistance against photobleaching. However, it has also become clear that the protein forms closely packed tetramers, and there is indication for incomplete protein maturation with unknown proportion of immature green species. We have ...

Autofluorescent proteins, in particular the well-known green fluorescent protein (GFP) have revolutionized the field of fluorescence imaging in molecular and cell biology, providing researchers with a direct and unique view into the dynamic organization and protein–protein interactions that occur in living cells. Spectrally distinct blue-shifted and red-shifted GFP mutants have been developed t...
